INS505: FREEZE-DRY MICROSCOPY (FORMERLY COM450)
Freeze-dry microscopy, as part of a complete thermal analysis study, is an invaluable tool in the characterization of the thermal properties of any formulation. It allows you to save a significant amount of time in money both in process development and in commercial manufacturing. Freeze-drying, as a unit operation in the production of parenteral products, is becoming more prevalent as many of the new molecular entities coming out of discovery do not have sufficient shelf life in the solution state (< 2 years). Since freeze-drying is the most expensive and time consuming step in the parenteral manufacturing process, it is in the best interest of companies developing new products to make sure that they are running an optimized lyophilization cycle.
Our course begins with presentation of some of the basic theories behind lyophilization and polarized light microscopy, followed by a “hands on” demonstration of the set up and alignment of the freeze-dry microscopy system. Each student practices using the freeze-dry microscope system by measuring the thermal properties of various crystalline and amorphous samples. The course finishes with a demonstration of how the collected data can be used for lyophilization cycle development and optimization.

- Principles, theory, and practice of freeze-dry microscopy using a polarized-light microscope (using an Olympus BX51 microscope)
- Anyone who is involved in the development, optimization, or troubleshooting of lyophilization cycles would benefit from this course. The course will be taught with a limited amount of theory which will be presented at a level that even someone new to the sciences of lyophilization and polarized light microscopy will be able to comprehend. Individual “hands on” training using the equipment will give students the tools necessary to take the skills they have learned in the course and apply them for their own lyophilization development or optimization needs.

Students are expected to successfully complete a variety of tasks in the form of hands-on exercises, laboratory exercises, identifications of unknowns, and quizzes. In addition, the students are required to have 100% attendance during the course, participate in class, complete a student evaluation form and pre and post course assessment forms.
The student is notified at the end of the course whether or not they have successfully completed the requirements of the course based on:
- 100 % attendance
- class participation
- completion of all course material
- completed and signed student evaluation form
- completed pre and post course assessment forms.
Upon successfully meeting these requirements, a student is awarded a certificate of completion and IACET CEU credits, if available. Those who have not successfully passed the course requirements do not receive a certificate or IACET CEU credits.
